Aging skin symptoms such as wrinkles, sagging, and a decrease in firmness are caused by the degradation of the extracellular matrix (ECM) such as collagen in the dermis. Dermis with a degraded ECM becomes thinner, making the skin look thin. According to recent studies, the root cause of skin thinning is considered to be extracellular ATP produced by epidermal cells.

Mechanism of extracellular ATP and skin thinning

As cells aged, the extracellular ATP release increases. ATP released into the extracellular environment is a factor that transmits stimulation. Aging cells stimulated by extracellular ATP exhibit the senescence-associated secretory phenotype (SASP), secrete SASP-related factors and induce aging in surrounding cells. Fibroblasts in the dermis receiving these SASP-related factors secrete ECM degrading enzymes such as MMP, destroying the dermal layer and causing skin thinning of the dermis.
